高转移人肺癌细胞系遗传稳定性研究
Study of genetic stablility of the highly metastaic human lung cancer cell line
【摘要】 染色体畸变分析为肿瘤细胞遗传学研究的常规方法。近来,有研究表明癌细胞长期培养并不会引起额外特征性染色体改变。PG 癌系为我室建立的具有高转移性人肺癌细胞系,在体外已传代数年。最近,我们从 PG母系中分离出多个亚克隆,并具有相同的生物学特性。为探讨其遗传稳定性及进一步研究分子遗传改变与生物学行为的关系,我们比较了 PG 母系与两个亚克隆的染色体畸变。结果表明:1,3,6,7,11,13,17,19号染色体异常与其它肺癌的研究报道相近;母系与亚系的染色体畸变类型几乎一致。因此说明 PG 细胞在长期培养过程中,其遗传特性是稳定的,并相似于一般肺癌遗传学改变。
【Abstract】 Analysis of chromosome abnormality has become the routine method in oncological cy- togenetics.However,it is considered that whether artifacts in chromosomal changes can be induced dur- ing in vitro cell culture,which is a necessary procedure in chromosome prepation.Recently,some re- ports indicated that long-term culture of cancer cells in vitro would not cause specific changes in chro- mosome.PG cell line is a highly metastatic human lung cancer cell line,and has been passed many years in vitro.Recently,a few subclones have been isolated from PG cell,which have identical biologi- cal behaviors.To probe the genetic stabilitly and get insight of the basis for the biological features,the chromosomal abnormalities of PG and its subclones were compared The results showed that abnormality in chromosome 1,3,6,7,11,13,17,19 were similar with those of other reports about lung cancers and the alterations in subclones and parent PG cells were no different.This suggested that the genetic fea- tures of PG cell were stable and shared similarity with lung cancers.
