抗毒灵注射液抗心律失常机制的电生理研究
Eiectrophysiologic Study on the Mechanism of Antiarrhythmic Effect of Kangduling(抗毒灵)Injection
【摘要】 本研究采用玻璃微电极技术观察了抗毒灵注射液对豚鼠乳头肌动作电位的影响,结果表明:(1)该药能明显减低0相最大去极化速度(Vmax),减慢2相复极化速率,使有效不应期/动作电位时程90(ERP/APD90)增大,延长动作电位时程和有效不应期,并通过维拉帕米阻断慢通道,该药仍能减慢2相复极化;(2)对儿茶酚胺诱发的高钾去极化心肌的慢反应动作电位,该药能减低 Vmax 及动作电位幅度。认为,该药抗心律失常作用与其抑制 Na+内流、Ca++内流及 K+外流有关。
【Abstract】 Kangduling injection is a Chinese herbal mixture.The results of some recent experiments on animals confirmed that it could be used to prevent or treat arrhythmias induced by digoxin in rabbits.In the light of these findings,the mecha- nism of its antiarrhythmic effect was studied with glass microelectrode.The results were as follows:(1) Kangduling injec- tion could significantly decrease Vmax (P<0.01),and lengthen the duration of PAD and ERP,the value of ERP/ APD90 became greater;It also postponed the repolarization in the plateau phase.When the slow sodium current and the slow channels were blocked by verapamil,Kangduling injcetion could postpone the reploarization in phase two.(2)The effects of Kangduling injection on the slow response potential induced by catecholamine in guinea pig papillary muscle de- polarized by high potassium were also observed.During the administration of the agent,the APA,Vmax of the slow po- tentials were increased(P<0.05).Above all,the antiarrhythmic effects of Kangduling injection were as follows:it could significantly inhibit the rapid sodium inward current,the slow calcium inward current and the potassium outward current.
