对硝基酚钠生理药物动力学模型的安全性评价
Estimation of Safety and Toxicity of ρ-Nitrophenol Sodium with a Physiologically Based Pharmacokinetics Model
【摘要】 利用生理药物动力学模型,对对硝基酚钠在大鼠及人体内的动力学进行研究,结果大鼠模型预报值与实验结果基本一致,人体模型中肾脏与肝脏的最高浓度预报值为大鼠模型的1.5和1.7倍。探讨了以生理药物动力学模型对化学物质进行安全性评价的可能性。
【Abstract】 The safety and toxicity of chemicals given first to animals and finally to humans are gen-erally estimated with a method of safe coefficient,which lacks scientific ground.To changethe traditional method,we designed a physiologically based pharmacokinetics model(PBPK)for the estimation of safety and toxicity of chemicals.For example,ρ-nitrophenol sodium(PNP-Na)was used.The PNP-Na pharmacokinetics in bodies of rat as well as humans werestudied,and possibilities of making use of the model in the estimation of safety and toxicity0fChemica15 were also discussed.
