大鼠内脏伤害性刺激后脑内70kDa热休克蛋白和c-Fos表达
EXPRESSION OF 70kDa HEAT SHOCK PROTEIN AND C-FOS LIKE PROTEIN IN RAT BRAIN AFTER VISCERAL NOCICEPTIVE STIMULATION
【摘要】 不同的内脏伤害性刺激动物模型上研究脑内c-fos样蛋白(Fos)表达的研究己有报道,而具有分子伴侣功能的70kD热休克蛋自(hsp70)的表达是否有改变尚未见报道。本工作以大鼠内脏大神经电刺激作为伤害性刺激动物模型用免疫组化方法观察脑内hsp700和Fos表达的变化。结果显示:在内脏大神经电刺激后12-18h于大脑新皮质,内嗅皮层,尾核,丘脑外侧核后部,下丘脑外侧核,下丘脑弓状核,巨细胞网状外侧核等结构观察到hsp70免疫阳性细胞;Fos在蓝斑,中脑导水管周围灰质,中缝大核,背核,下丘脑弓状核,隔核,伏隔核,新皮质等部位于刺激后2h表达增加,4h后减少。对照组脑内hsp70和fos阳性染色很少。这提示:hsp70和Fos表达可能都参与神经系统对内脏伤害性刺激的反应并发挥不同的功能。
【Abstract】 Although induction of the c-fos like protein (Fos) has been demonstrated in brain following different kind of experimental noxious stimulation, whether the 70 kDa heat shock protein (hsp70) could be induced with the same stimulation is still unknown. In the present study, hsp70 and Fos was localized in rat brain at intervals after visceral nociceptive stimulation by using immunohistochemical method. Induced hsp70immunoreactivity was found only in neocortical regions, entorhinal cortex, caudats putamen, neucleus lateralis thalami pars posterior, nucleus ventromedialis hypothalami, nucleus lateralis paragigantocellular, hypothalamic arcuatus at 12-18 h after electrical stimulation of greater spanchnic nerve (GSN). The fos immunoreactivity was present at 2 h, declined after 4 h in locus coeruleus, periaqueductal gray, raphe dorsilis, raphe magnus, nencleus reunieus-rhomboid thalamic nucleus, hypothalamic arcuatus, dorsomedial hypothalamic nucleus, sepia lateralis,accumbens and neocortical reigon. But in the control rats, little expression of both hsp70 and Fos was observed.It is suggested that Fos and hsp70 induction are both components of neuronal response to visceral nociceptive stimulation and may take different functional roles.
