木聚糖降解菌的筛选和木聚糖酶性质的研究

【摘要】 筛选两株生长快、产木聚糖酶活力高的菌种;黑曲霉(Aspergillusniger,AN01)、链霉菌(Streptomycessp.Str7B),酶活力分别达128mg/L·min和176mg/L·min;酶反应最适温度分别为60℃与50℃;最适pH值为5.0和6.0,并分别在pH2.2~5.0,5.8~6.4酶活性稳定;在60℃条件下保温1.5h,酶活力分别剩余20.5%,88.5%,其中AN01株原酶液在90℃保温10min,活力仍剩余14.5%.Cu2+对酶活表现出极强抑制,Fe2+,Mg2+,Ca2+等离子则有促进作用;用纸层析法探讨了不同培养时间各种产物产生的情况.

【Abstract】 Two strains of Aspergillus niger (AN01) and Streptomyces sp. (Str7B) with fast growing and high xylanase activity were isolated from soil samples. The enzyme activity of AN01 was 128 mg/L. min, at 30℃ for 72h. with shaking and that of Sir7B was 176mg/L. min, at 50℃ for 72h. The optimal temperature and pH for xylanase reaction were 60℃ and 5.0 for AN01 and 50C, 6. 0 for Sir7B. The enzyme activity of AN01 was stable within PH2. 2-5.0 and that of Sir7B was PHS. 8-6.4. The remaining activity of AN01 and Sir7B xylanase were 20. 5% and 88. 5% after incubation at 60℃ for 1.5h, Fe2+, Mg2+ and Ca2+ increased both of the activities whereas Cu2+ inhibited them strongely. The products of enzyme hydrolysis in different culture times shown bymeans of paper chromatography were also discussed.
