Tagging of a gene for aroma in rice by RAPD and RFLP (Ⅱ)
【摘要】 用RAPD引物Jas15从亲本CT9993中扩增出的多态性产物15kb片段,经NcoⅠ酶切后获得的一条05kb片段,将其制备成RFLP探针jas500。选用覆盖整个水稻RFLP图谱的125个随机克隆和jas500对CT9993/KDML105的F2群体进行检测,结果表明:KDML105的香味遗传受一对隐性基因控制;控制香味基因aro与jas500以及第8染色体上的几个RFLP标记有连锁关系,并将其初步定位于第8染色体上jas500和C222之间,距离分别为158cM和278cM,该香味基因在RFLP图谱上的初步定位为进行该基因的精细定位及有效利用奠定了基础,RFLP标记jas500和RAPD引物Jas15均可直接应用于水稻育种实践。
【Abstract】 A 0 5 kb fragment was obtained by digesting with restriction enzyme NcoⅠ from the 1 5 kb band amplified by RAPD primer Jas 1 5 in CT9993 and developed into an RFLP marker named jas 500 Aromatic evaluation in the progenies of the cross indicated the presence of a major gene in KDML 105 A recessive locus for jasmine-like aroma in KDML 105 was detected and mapped between RFLP markers jas 500 and C222 on chromosome 8 The distance between jas 500 and aro gene was estimated to be 15 8 cM, and C222 is on the other side of aro gene at a distance of 27 8 cM Closely linked RFLP marker jas 500 as well as RAPD marker Jas 1 5 may facilitate early selection for aroma gene in rice breeding programs.
