人PBL及CTLL-2/HIL-4R细胞系对人IL-4增殖应答的特性

【摘要】 本文分析了30例正常人PRL经PHA活化的T细胞对IL-2及IL-4的增殖应答特点;及整合有人IL-4受体cDNA的CTLL-2/HIL-4R细胞系对人IL-4的特异增殖应答特点。结果表明,正常人经PHA活化的PBL对IL-2有良好增殖应答曲线者占93%,而对IL-4有良好增殖应答者只占20%。 CTLL-2/HIL-4R细胞系对人IL-4呈规律性增殖应答曲线,且应答敏感性高,可测出0.09u/ml,可推荐作为测定人IL-4生物活性的一种准确方法。

【Abstract】 Human Interleukin 4 (hlL-4 ) is a cytokine with important immuno-regulating activities. Precise assessment of hlL-4 activity become. increasingly critical. We reported the characteristics of proliferative response of 30 samples of PHA activated T cells in PBL obtained from normal human donors to rhlL-2 and rhlL-4. The hlL-4 bioactivity was also assessed by its support of the proliferation of CTLL-2/HIL - 4R cell line ,a CTLL-2 derived cell line integrated with hlL -4 receptor CcN A . Through statistical analysis , our results showed that 93% of the samples of PHA activated PBL responded well to IL -2 by proliferation with a positive correlation of dose response. In contrast ,only 20% of these samples showed good dose-response curve to hlL-4 ,The sensitivity of IL-2 and IL -4 is 0. 5U /ml and 8. 2U /ml respectively. As expected , CTLL- 2/HIL-4R cell line exhibited susceptible to hlL-4 at a dose as low as 0. 09U /ml and regular dose-response curve. It is , threfore , recommendatory that CTLL-2/HIL-4R cell line could be adopted to accturately assess hlL - 4 bioactivity.
