人源化IL-2免疫毒素的构建和鉴定
Construction and identification of a humanized IL-2 immunotoxin
【摘要】 利用基因工程技术在大肠杆菌表达成功人白细胞介素2(IL-2)和人血管生成素(Angiogenin)的融合蛋白(IL-2-Ang)。活性检测证明,它具有IL-2免疫毒素的作用,在体外可以杀伤IL-2依赖性CTLL细胞株和经ConA活化的小鼠脾脏IL-2R阳性细胞。在双相混合淋巴细胞培养试验中,IL-2-Ang可抑制淋巴细胞的增殖。在体内实验中,IL-2-Ang注射小鼠可使其抗原活化的脾细胞数量减少。由于IL-2和Ang均为正常人体成分,IL-2-Ang融合分子作为人源化免疫毒素可望具有更好的应用前景。
【Abstract】 A novel fusion protein namely IL- 2-Ang consisting of interleukin-2 ( IL-2 ) and angiogenin ( Ang) was expressed in E. coli by using genetic engineering technique. Bioassays showed that it could be used as an IL-2 immunotoxin. In vitro ,the IL-2-Ang exhibited cytotoxity effects on the IL-2-dependent cell line CTLL as well as murine IL-2R positive splencytes activated by ConA. In vivo, injections of IL-2-Ang into mice could decrease the splencytes activated by antigen. Since both of the IL- 2 and Ang originated from human beings , the IL-2-Ang could be used as new generation of humanized IL-2 immunotoxin.
