NB4细胞生物学特性与维甲酸诱导分化机理研究
Blological Properties of NB4 Cell Line and Differentiating Induction of Retinoic Acid
【摘要】 研究了NB4细胞多项生物学参数及三种维甲酸(RA)对NB4细胞增殖分化的影响,以探求RA诱导分化的机理。形态学观察、细胞生长曲线、DNA含量分析、Brda掺入、膜分化抗原测定、硝基蓝四唑(NBT)试验等研究表明,NB4细胞DI=1.73,TD=24.9小时,Tc、Ts、T(??)分别为5.1、15.4、4.4小时;NB4细胞诱导分化过程中,前48小时伴有明显的细胞增殖;RA诱导分化作用不表现细胞周期特异性。在浓度为10-8mol/L下,三种RA的效应强度为9c-RA>ATRA>13c-RA。实验结果对阐明RA诱导早幼粒白血病细胞分化的作用机理,解释其临床疗效与副反应有一定价值,并对评价新型RA提供依据。
【Abstract】 Several biological properties of NB4 cells and effect of three isomers of retinoic acid (RA) on NB4 cells have been studied for the mechanizm of differentiation induction of RA. The observation of morphological changes, cell growth curve, DNA content analysis, cell BrdU incorperation, cell membrane antigen determination, nitro blue tetrazolium (NET) test indicated that the DNA Index (DI) of NB4 cells is 1. 73, its doubiling time (To) is 24. 9 hours. TGI , Ts and TGZM are 5. 1, 15. 4, 4. 4 hours respectively. During the first 48 hours of RA treatment, the "NB4 cell differentiation was coupled with cell proliferation. The effect of RA has no cell cycle specificity. At the concentration of 10-8mol/L2 the effect strength order of three RA isomers is 9C-RA>ATRA>13c-RA. These results provided some clues to explain the mechanizm of RA differentiation induction and some clinical side effects, and showed that 9C-RA could be a.promising drug.
