食管癌ras癌基因蛋白p21、抑癌基因蛋白p53和DNA的定量研究

【摘要】 定量分析食管癌细胞ras p21、p53蛋白表达和DI、PI对食管癌早期诊断、预后的作用。采用FCM,应用单克隆抗体对50例食管鳞状细胞癌组织及39例残端组织石蜡包埋标本定量检测显示,食管癌Tas p21、p53平均表达量、DI和DI均明显高于正常食管(P<0.01)。淋巴结转移阳性组与阴性组p53蛋白高表达率分别为83.33%、69.23%(P>0.05)。39例残端病理证实为阴性、不典型增生及原位癌中,ras p21、p53蛋白表达及PI依次呈增高趋势。四项指标均增高提示食管癌具有恶性地浸润生长活性。p53蛋白高表达与淋巴结转移有关。残端ras p21、p53蛋白表达和DI三项指标的升高提示残端组织细胞增生活跃。

【Abstract】 For early diagnosis of esophageal carcinoma and evaluation of the prognosis, ras p21, p53 protein, DNA index (DI), proliferative index (PI) were quantitatively studied. Tissue blocks of paraffin-em-beded material from 50 patients with esophageal squamous cell carcinomas (ESCC) and 39 corresponding esophageal resection line were detected by flow cytometry. The results showed that the mean expression quantity of ras p21, p53, DI and PI was significantly higher than that of normal esophageal mucosa specimens (P<0.01). Overexpressing p53 protein (FI>1.10) was 83.30% in lymph node involvement group and 69. 23% in no lymhp node involvement group (P>0.05). The ras p21, p53 protein, DI, PI were analysed from resection lines of cases of ESCC. In the sample of residual negative, atypical hyperplasia and carcinoma in situ, the level of ras p21, p53 protein, DI were grandually increased. The increased values of all four indexes indicated a higher growth activity of ESCC. Overexpressing p53 was related to metastasis of lymph node. The increases of residual ras p21, p53 protein and DI indicated an active proliferation of residual tissues.
