二膦酸盐HEDP和HPDP治疗去势大鼠骨质疏松症的实验研究

【摘要】 用作者合成的二膦酸盐HEDP(1-羟基乙叉二膦酸二钠)和HPDP(1-羟基戊叉二膦酸二钠)治疗去势雌性SD大鼠骨质疏松症,采用椎骨纵向压缩试验、生化测定和形态学观察等方法,探讨了HEDP和HPDP对去势大鼠的治疗作用及其对骨矿化的影响。结果表明:HEDP(2 mg/kg),HPDP(2,8 mg/kg)能治疗去势大鼠的骨质疏松;直接抑制破骨细胞的功能是治疗骨质疏松的机理之一。HEDP和HPDP两者的实验结果比较说明,恰当延长HEDP结构中的侧链基团可望增强疗效,减轻对骨矿化的抑制作用。

【Abstract】 The ovariectomized SD rats were treated with HPDP(1-hydroxyethylidene-1,1-diphosphohate) and H EDP (1-hydroxypentylidene-1,1-diphosphonate) respectively which were synthesized in our laboratory. The longitudinal compressing experiment, mophological observation and biochemical assay were carried out to evaluate the effects of treatment of osteoporosis with diphosphonates and its influence on bone mineralization. The resutls showed that HEDP(2 mg/kg) and HPDP(2,8 mg/kg) were effective on treatment of osteoporosis induced by ovariectomy on rats. It indicated that the function of osteroclast could be inhibited when osteoporosis was treated with diphosphonates. Comparing the experimental results of the action of HEDP and HPDP, it also indicated that suitable prolonging the side chain of HEDP might improve its efficiencies and diminish the inhibition of bone mineralization.
