【摘要】 本研究以１１．８ＫＤｈＢＭＰ为抗原，通过脾内直接免疫法对ＢＡＬＢ／ｃ小鼠进行免疫。用３０％ＰＥＧ１５００对ＳＰ２／０骨髓瘤细胞：免疫脾细胞（１∶５）的混合细胞悬液进行融合。融合后将２４孔板中每孔种植含１×１０６个细胞的混合细胞悬液，可使融合率达１００％。通过筛选，从２５００株杂交瘤中选出５株杂交瘤，它们能产生特异性较好的抗１１．８ＫＤｈＢＭＰ的单克隆抗体。用抗１１．８ＫＤｈＢＭＰ单抗对颈椎骨赘、项韧带异位骨化标本、滑膜软骨瘤病的瘤结节进行ＡＢＣ免疫组化染色。结果显示，颈椎项韧带异位骨化、颈椎椎体骨赘形成及滑膜软骨瘤病的发病与ＢＭＰ的异常表达有关。更多还原

【Abstract】 BALB/c mice were immunized by injecting 11.8KD hBMP entraspleen.Cell fusion was done by using 30% PEG.It had been proved that when the myoloma cells:imm unized spleen cells was 1:5,the fusion rate would be 100%if 1×10￣6 cells had been implanted into each well of Linbro 24-well plate.Five lines of hybridomas which secreted anti 11.8KD hBMP McAb with high specialization were selected from 2500 lines of hybridomas.Anti 11.8KD hBMP McAb was used to inspect the cervical vertebral osteophyte,the nuchal ligament’s ectopic ossification and the tumour node of synovial chondromatosis with ABC immunohistochemical stained method.The result showed that the ectopic ossification of nuchal ligament,the formation of cervical vertebral osteophyte and the pathogenesis of synovial chondromatosis were related to unusual expression of BMP.更多还原
