环孢霉素A对游离肝细胞核酸及蛋白质合成的影响

【摘要】 用SD大鼠肝组织原代培养研究了环孢霉素A对肝细胞蛋白质及核酸合成的影响。不同浓度的环孢霉素A加入培养液,培养4h后加3HTdR,3HUrd,3HLeu入培养液继续培养12,24及36h,测肝细胞放射性强度。发现环孢霉素A可抑制RNA、蛋白质合成,对DNA无明显影响。

【Abstract】 The effects of cyclosporine A(CsA) on the syntheses of DNA, RNA and protein were evaluated by the method of 3 H labelled TdR, Urd and Leu in rat primary hepatocyte culture. Various doses of CsA were added into the cultures. After incubation for 4 hours, 3 H TdR, 3 H Urd and 3 H Leu were added into the mediums. The hepatocytes were cultured for another 12, 24 and 36h respectively, and then the cpm in the hepatocytes were assayed. The results showed that CsA could inhibit the syntheses of RNA and protein, but had no significant effect on the DNA synthesis of hepatocytes.
