白血病细胞内SOD及GSH-Px活性研究
Study on Superoxide Dismutase and Glutathione Peroxidase of Leukemic Celis
【摘要】 观察了29例白血病患者白血病细胞内超氧化物歧化酶(SOD),谷胱甘肽过氧化酶(GSH—Px)活性及血清脂质过氧化物(LPO)水平。研究结果表明:①白血病细胞内SOD活性明显升高,GSH—Px活性显著降低,血清LPO水平增高;②白血病细胞内SOD及GSH—Px活性分别与外周血白血病细胞百分数呈正负相关;③与单核细胞白血病(M4,M5)及淋巴细胞白血病相比,粒细胞白血病(M1,M2,M3)细胞内SOD活性相对较高,GSH—Px活性相对较低;④白血病细胞内SOD活性在白血病缓解与复发过程中无显著性差异,而GSH—Px活性则在缓解后回升,复发时又重新降低。上述结果提示,白血病细胞自身存在有明显的过氧化物损伤,给予白血病患者抗氧化治疗是必要的,白血病细胞GSH—Px活性可作为白血病活动的观察指标。
【Abstract】 This paper presents an analysis of superoxide dismutase (SOD), glutathione peroxidase (GSH-Px) activities of leukemic celis in 29 patients with acute leukemias and serum lipid peroxides (LPO) Ievel. Blast celis were isolated from peripheral blood before induction therapy using Ficoll density gra-dient. SOD activity was measured by a cytochrome C reduction method. GSH- Px activity was mea-sured by Levander’ s method. Results show: ① Higher SOD activity and lower GSH - Px activity were found in leukemic celis and serum LPO increases in leukemic patients; ② The positive correlation between SOD activity and perpheral blast cell counts and the negative correlation between GSH - Px activity and blast cell counts were obtained; ③ According to FAB classification, the highest SOD ac-tivity and lowest GSH - Px activity were found in the myeloblastic leukemia (M1, M2 and M3); ④ No significant difference of SOD activity was observed between remission and relapse of leukemia. Howev-er, significant diference of GSH - Px activity was found in the course of remission and relapse in leukemia. These results suggest that: ① leukemic celis, especially FAB- M3 blastic cells, are exposed to injury by lipid peroxides. It is necessary that patients with leukemia accept antioxidant treatment; ② GSH-Px (not SOD) activity of leukemic celis could be a favourable factor in activity of the dis-ease.
