对脐血CD34~+CD38~-细胞的研究揭示脐血移植成人的可能性

【摘要】 通过在生长因子依赖的长期培养体系中加入转化生长因子β1(TGF-β1)的反义核酸或抗TGF-β1抗体以阻断TGF-β1的抑制作用,比较了脐血和骨髓的CD34~+CD38~-及CD34~+CD38~+亚群在长期培养中的增殖反应以及产生早期造血前体细胞的能力。结果显示:脐血中CD34~+CD38~-细胞占CD34~+的比例为4%,比骨髓多4倍;和成人骨髓移植平均所需的骨髓标本相比,一份常规脐血标本含有的CD34~+CD38~-细胞所产生的CFU-GEMM和骨髓相当,CFU-GM为骨髓的2倍,BFU-E为骨髓的3倍;由脐血产生的集落明显大于由骨髓产生的集落。提示脐血更富含造血干细胞并具有较高的增殖潜能。脐血中晚期祖细胞量低于骨髓。以上研究结果提示脐血可用于成人进行造血细胞移植。

【Abstract】 Using cytokines-dependent long-term culture system in which the inhibitory effect of TGF-β1 has been blocked by antisense TGF-β1 oligonucleotides or anti-TGF-β1 serum, the proliferative ca-pacities and the abilities of the CD34+CD38-and CD34+CD38+ cell population to generated early progenitors form umbilical cord blood have been compared with that from bone marrow. The CD34+CD38- population in CD34+ fraction from umbilical cord blood is 4 times more than that from bone marrow. We estimate that the CD34+CD38 celis from a typical umbilical cord blood sample produce equivalent CFU-GEMM, twice as many CFU-GM and 3 times as many BFU-E as the same population from an average bone marrow sample for adult transplantation. In addition, the colonies derived from umbilical cord blood were significantly larger than those from bone marrow. These results indicate that umbilical cord blood sample may be relatively enriched in stem celis and have a greater growth potential. However, the content of later progenitors was less in umbilical cord blood-derived than in bone marrow-derived cell populations. These studies suggest that umbilical cord blood is a suitable source of cells for adult transplantation.
