金樱子果肉Cu·Zn—SOD纯化及其活性测定

【摘要】 通过(NH4)2SO4分级沉淀和SephadexG—100分离提纯得到金樱子Cu·Zn—SOD,经聚丙烯酞胺凝胶电泳发现有5—6条同工酶谱带,最明显的3条迁移率分别为0.43,0.46和0.55,SDS—PAGE测得亚基分子量约为13KD,其活性被2mmol/LKCN或H2O2抑制,不受氯仿—乙醇影响,在pH7.6磷酸缓冲液中活性最大,对热相对稳定。SOD活性与VC、GSH含量有明显的相关性,相关系数为0.9342和0.9106,果实由金黄转褐黄阶段有—SOD活性高峰。认为以Vc为指标可以判断SOD活性水平。

【Abstract】 Superoxide dismutase (SOD) from fruit of Rose laevigata has been purified with the fractional percipitation by ammonium sulfate and separation by sephadex G100. The enzyme subumit is about 13KD as assayed by SDS PAGE. The SOD was relatively heat stable and there was a higher activity at PH7. 6. The SOD activity was found to be inhibited by 2mmol/L KCN or 2mmol/L H2O3,but was not affected by chloroform ethanol. When the crude extract of Rosa laevigata SOD was passed through polyaerylamide gel during electrophoresis and stained for enzymic activity with NBT, it was shown to contain five or six distinguishable transparent isoemyme bands or a blue background, migrating rate was 0.43, 0.46 and 0.55 respectively. The color of Rosa laevigata coat changed from gold yellow to red with a maximel SOD activity peak.There was a positive correlation between the SOD activity and the contents of Vc and GSH, Which correlation coefficient was 0.9342 and 0.9106 respectively. We propose that the Vc content may distinguish the SOD activity.
