荧光光谱法研究四环素族化合物与白蛋白的相互作用

【摘要】 用荧光光谱法研究了四环素族药物(四环素,土霉素,金霉素)对白蛋白荧光的猝灭.由静态猝灭法和Scatchard法获得了药物与白蛋白作用的解离常数,且与其它方法获得的结果相符.说明它们的作用过程主要是静态猝灭.鉴于白蛋白与四环素族化合物之间可以发生能量转移,可通过Foster偶极-偶极无辐射能量转接原理,测得白蛋白氨基酸残基与四环素族药物之间临界距离R0,以及在它们各自能量转移效率情况下,白蛋白荧光团与四环素族分子中心距离r.由此提出形成非发光复合物模型

【Abstract】 The quenching fluorescence of albumin by tetracyclines was studied by fluorescence Spectroscopy.The dissociation constants between tetracyclinesmedicine with HSA or BSA were obtained according accorded to static quenching and Scatchard equation.These results accorded with those from the other methods,and showed that the interaction process was static quenching.By use of a spectra overlap intergral between the absorption spectrum of these compounds and the emission spectrum for HSA or BSA,the distance of Trp residue to these compounds was estimated.In addition,the binding mechanism was discussed in brief and an model of a tetracyclines HSA or a tetracyclines BSA non fluorescent complex was pcstulated from above results.
