硒多糖、亚硒酸钠对大鼠肝线粒体酶的影响及其拮抗亚砷酸钠毒性作用的机理
The Antagonism and Mechanism of Selenium Compounds against Sodium Arsenite Inhibition Effect on Mitochondrial Enzymes in Rats
【摘要】 采用体内、体外实验方法研究了硒化合物(硒多糖、亚硒酸钠)和亚砷酸钠对大鼠肝线粒体丙酮酸脱氢酶、琥珀酸脱氢酶、谷胱甘肽过氧化物酶的活性以及对肝线粒体膜脂质过氧化、还原型谷胱甘肽含量的影响,探讨了其作用机理。结果表明:硒化合物可明显拮抗亚砷酸钠对线粒体丙酮酸脱氢酶、琥珀酸脱氢酶活性的抑制作用,且硒多糖的抑制作用较亚硒酸钠强。这种拮抗作用在防治砷中毒方面具有重要意义。
【Abstract】 The effects of selenium compounds(selenium polysaccharide and sodium selenite)and sodium arsenite on pyruvate dehydrogenase(PDH) and succinate dehydrogenase (SDH)of mitochondria in rats were studied through in vivo and in vitro experiments,and the mechanism was also illuminated preliminarily by determining their effects on the activity of glutathione peroxidase (GSH-Px),the lipid peroxidation and the level of liver reduced glutathione.The results indicated:Sodium arsenite inhibited the activities of PDH and SDH of mitochondria in vivo and in vitro(P<0.05).The selenium compounds,which had no marked influence on the two enzymes,antagonized siginificantly the inhibiting effects of sodium arsenite on the activities of PDH and SDH. These findings suggest that selenium compounds can protect against the deleterious effects of arsenic on mitochondrial enzymes,and the effect of selenium polysaccharide is stronger than that of sodium selenite.
