大鼠卵巢内皮素-1受体的动态变化
Changes of endothelin 1 receptors in different stages of rat ovaries
【摘要】 应用放射配基结合分析法(RBA)检测了大鼠不同发育阶段卵巢内皮素-1(ET-1)受体的动态变化。结果表明:大鼠卵巢上存在有特异性ET-1受体,受体最大结合量(Bmax)于窦前期卵泡含量最高(358.2±73.2fmol/mg蛋白),随卵泡发育到排卵前卵泡时显著降低(162.7±17.2fmol/mg蛋白)(P<0.01),排卵后的早期黄体化卵巢降至最低值(37.8±3.4fmol/mg蛋白)(P<0.01),于晚期黄体化卵巢又再次升高(95.3±4.9fmol/mg蛋白)(P<0.01)。受体亲和力(Kd值)也随卵泡和黄体发育的不同阶段呈现动态变化,在排卵前卵泡卵巢ET受体对ET-1的亲和力最低,晚期黄体化卵巢最高。结果提示,ET-1可能通过与ET-1受体结合参与卵泡和黄体发育的调节。本结果为ET-1作为卵巢内局部调节肽提供实验依据。
【Abstract】 Objective: To determine the changes of endothelin 1(ET 1) receptors in ovaries containing preantral follicles,ovaries containing preovulating follicles,early luteinized ovaries and late luteinized ovaries of rats. Design: Labortory studies. Setting: Laboratory in Department of Physiology,Jiangxi Medical College. Methods: Preparations of ovarian membrane from ovaries in different stages.Samples were used for radioligand binding assay of receptor. Results: Radioligand binding assay of receptor revealed that endothelin 1 specific receptors were present in rat ovaries.The maximal binding capacity(Bmax) varied with follicles developing and corpus luteum(CL) degenerating.Bmax was higher in ovaries containing preantral follicles than in ovaries containing preovulating follicles(358.2±73.2 and 162.7±17.2 fmol/mg protein,respectively) ( P <0.01) After ovulating Bmax varied with CL age,namely,higher in old CL than in young CL(95.3±4.9 and 37.8±3.4 fmol/mg protein,respectively)( P <0.01).The affinity (Ka) also varied with follicles developing and CL degenerating.Dissociation constant values (Kd) were 6.2×10 -11 ,9.5×10 -11 ,3.5×10 -11 ,3.1×10 -11 mol/L in the ovaries containing preantral follicles,preovulating follicles,early and late luteinized ovaries,respectively.Ka was lowest in the ovaries containing preovulating follicles and highest in late luteinized ovaries. Conclusion:The study suggested that ET 1 may be an intra ovarian factor which regulates follicular development and corpus luteum degeneration through endothelin 1 specific receptors.
