人尿集落刺激因子的分离及其特性研究
Primary Separation and Purification of Colony—Stimulating Factor(CSF)from Human Urine
【摘要】 本实验采用(W/V)K—9型高岭土吸附、CM—52柱层析及 DE—32柱层析技术,对正常人尿集落刺激因子(CSF)进行分离、纯化.结果表明:(1)用人尿提取 CSF,取材简便,来源丰富,制备容易,是天然 CSF 的可靠来源;(2)人尿 CSF 有两种不同的蛋白,它们确能刺激小鼠骨髓细胞形成CFU—GM;(3)经典的集落培养法测定其活性更靠便可靠。
【Abstract】 The method of separation and purification of CSF from normal human urine using a koli adheren- ce,CM—52 and DE—32 chromatography techniques is reported.The results show that:(1)The human urine is rich in CSF and it is a reliable resource of nature CSF;(2)Human urinary CSF(HU— CSF)comprises two kinds of proteins,with different PI and Molecular Weight:Their effects are species specific.Both of them could stimulate the progenitor cells of mice bone marrow to form CFU—GM; (3)CSF activity microassay by colony formation in agar measurment is much easier and morereli- able,while MTT measurement is more rapid.
