杀虫抗生素阿佛米丁紫外分光光度测定法
UV SPECTROPHOTOMETRIC DETERMINATION OF THE INSECTICIDAL ANTIBIOTIC AVERMECTIN
【摘要】 大环内酯类杀虫抗生素阿佛米丁及其同系物,在紫外246um处有一吸收峰。由有机溶剂抽提的阿佛米丁粗提物在60℃水浴蒸干溶剂,油状物用少量硅胶吸附,加水离心洗涤去除色素和杂质,用甲醇洗脱后在紫外分光光度计测246um处光密度。在25~150μg/mL浓度范围内,阿佛米了浓度与A246呈良好的线性关系,其相关系数为0.9999。该方法可靠、精确,重复测定的变异系数在4.47%~7.68%,回收率为93.5%.该法测定结果与用生物测定所作的比较试验结果相符。该法操作简单、快速,无需昂贵设备,可用于高产菌株的选育和发酵工艺的控制。
【Abstract】 The insecticidal antibiotics avermectin and its homologs which have been identified as a series of macrocyclic lactone derivatives have an uv maximum at 246 um. The crude avermectins extracted with organic solvent were dried on a water bath at 60℃ to remove the solvent thoroughly, then the oily residue was adsorbed with a little of silica geland washed with water to remove pigment and other substance by centrifugation. Theavermectin complex was eluted with methanol and measured the A246 on an uv spectrometer. An excellent linear relationship was obtained between the A246 and the concentration of avermectin complex(25 ~ 150 μg/mL),the correlation index was 0. 999 9. The method described is reliable and accurate, The coefficients of variation were 4.47%~ 7. 65 % and the recovery rate was 93. 5 %. Comparative experiment with bioassay showed that the results agreed with those obtained by bioassay. The manipulation is simple, rapid and requires less expensive equipment. It can be used for selection of high-producing isolates and manipulation of fermentation process.
