应用免疫吸附电镜检测甘薯羽状斑驳病毒
Detection of Sweet Potato Feathery MottleViruses by Immunosorbent Electron Microscopy
【摘要】 应用血清学特异性的免疫吸附电镜法对接种SPFMV的I.setosa、I.nil和网室种植的感染SPFMV的徐薯-18、新大紫甘薯叶片分别进行了测定.结果表明被检测的四种植物汁液的稀释度分别可达到1/1280、1/640、1/640和1/160;而用无血清包被的电镜铜网检测I.setosa、I.nil和徐薯-18,其汁液稀释度仅达到1/20.比较接种SPFMV的I.setosa和I.nil植物体内病毒含量,前者高于后者.网室种植的自然感染SPFMV的两种甘薯品种,体内病毒含量也不一样.
【Abstract】 Sweet potato feathery mottle virus in I. setosa、I. nil、sweet potato of Xushu-18 and XinDaziwere detected by immunosorbent electron microscopy. The results show that SPFMV particleswere detected in inoculated I、setosa、I. nil and natural infected Xushu-18 and XinDazhi by ISEMat dilutions of 1/1280、1/640、1/640 and 1/160 respectively.Sap dilution of 1/20 was obtained byconventional electron microscopy for detecting SPFMV particles in I. setosa、I. nil and Xushu 18. Virus yeild was higher in leaves of I. setosa than in leaves of I. nil. It was different too inleaves of Xushu-18 and Xin Dazhi planted in greenhouse.
