环孢素A体外抗曼氏血吸虫幼虫的电镜学研究

【摘要】 MFI小鼠人工感染曼氏血吸虫3周后,经主动脉和门静脉灌注收虫.幼虫暴露在含不同剂量环孢素A的199型培养液中体外培养.药物所致的虫体损害作扫描电镜观察.药物作用24小时后,幼虫口吸盘出现肿胀、结构变形;外皮层显著水肿;进行性表皮损害和口吸盘闭锁,最终导致幼虫死亡.本结果表明,环孢素A具有直接杀曼氏血吸虫幼虫的作用,而口吸盘外皮质似乎是药物攻击的靶结构.

【Abstract】 MF1 mice were infected with Schistosoma mansoni cercariae for 3 weeks when me larvae were collected by aortic irrigation. Larval Schistsoma mansoni were exposed to varying quantities of Cyclosporin A in 199 culture medium for 24 hours in vitro.Drug in-duced damage to the worms was monitored throughout these experiments by SEM. Progressive surface damage,swelling of oral suckers and disorganization led to eventual death of larvae. These data support the view that the effect of Cyclosporin A against Schistosoma mansoni is direct and the oral suckers and tegument appears to be first focus for drug attack.
