一种新的HLA─Ⅱ类基因分型方法——PCR/SSP技术的建立
The establishment of PCR/SSP:a novel technique for HLA class Ⅱ DNA typing
【摘要】 PCR/SSP(Sequencespecificprimer,序列特异性引物)是一种新的基因多态性分析技术,可用于HLA复合体及其他具有多态性特点的基因分型。我室于1993年建立这种方法,并对正常人、重症肌无力患者、全身性红斑狼疮患者、骨髓移植供/受者进行了HLA-DRB1、-DQB1基因型别分析。与其它HLA基因分型方法相比,PCR/SSP具有简便、快速、准确及重复性好等特点。本文介绍PCR/SSP技术的原理、方法、特点以及常见问题的处理。
【Abstract】 PCR amplification with sequence specific primers(PCR/SSP) is a new method used to determine the allelic polymorphism of HLA chass Ⅱ molecules.After the establishment of this novel technique in our workgroup in 1993,HLA DBR1 typing using PCR/SSP were carried out among normal Hubei Chinese Hans,patients with myathemia gravis(MG)and systemic lupus erythematosus(SLE),donors and recipients of organ transplantation.It is demonstrated that the HLA DRB1 typing by PCR/SSP is simple,rapid and acurate,and is characterized with high resolution and good reproducibility,compared with other HLA class Ⅱ typing techniques such as PCR/SSO and PCR/RFLP.The principle,method,charactors of PCR/SSP as well as common trouble shootings were presented in this paper.
