免疫印迹实验及麻风菌抗原血清学活性的初步研究

【摘要】 初步分析了自己建立的聚丙烯酰胺凝胶电泳(SDS—PAGE)与免疫印迹试验(WB)及其应用于麻风菌(ML)和有关分支菌(AFB)抗原分析的结果,表明受试各菌都有自己独特的SDS—PAGE谱和WB反应谱。ML的WB反应带与有关报导大多相符,虽未能显示28KD,但发现ML有新的55KD反应带。作者认为,这些结果及特性可用于菌株鉴别和筛选血清试验用的吸收剂,对分析和寻找用于血清试验和疫苗的抗原及分离提纯这些抗原有相当的潜力。作者对实验方法及应用技巧作了详细描述,分析麻风菌(ML)抗原及其免疫活性对了解ML的抗原组成,探索麻风诊断、分型乃至发病机理都有重要意义。对此,国外已有些报道,但国内尚未见。

【Abstract】 SDS - polyacrylamide electrophoresis (SDS - PAGE) and Western Blotting (WB) based on SDS - PAGE were established, and used for analyzing and characterising the antigenic components of M. leprae (ML) and some mycobacteria (AFB) and their serological activities by using leprosy patient’s sera, This technique involved the separation of sonicates from ML and AFEs on SDS- PAGE,and the separated bands were eiectropheretically transferred to nitrocellulose (NC) and incubated the NC with patient’s sera. The visualization of the antigen-an-body complexes, was aoxie by indirect immunoperoxidase technique. The preliminary results showed that ML and the AFBs used in the test all have charateristic separation bands’ spectrum on SDS-PAGE and their sum of the bands were different,and that the positive reactional bands of the ML and AFBs were all showed on WB but no cross -reactiona! band between them. The sum of positive bands in ML were significantly more than other known AFBs, which were 70KD, 68KD,65,55,42/43,36,22, 18, 14 and 10KD and all have potential for immunodiagnosis. The 9ptimum conditions for use of SDS-PAGE and WB jointly or separately,purification,identification of ML and AFBs as well as for selecting absorbents of serological tests and so on were all discussed in detail.
