GUS与天花粉蛋白基因由花粉管通道导入小麦的研究
INTRODUCTION OF GUS AND TCS GNES INTO WHEAT VIA THE POLLEN--TUBE PATHWAY
【摘要】 切除羽状柱头后,在小麦花柱切面上滴注含有GUS与天花粉蛋白基因的质粒DNA。共处理小花1495朵,结种子1194粒,结实率为79.9%。随机取收获的烟农15种子125粒,室内萌发,PCR扩增证明有3株是含天花粉蛋白基因的转基因植株,转化率为2.4%。GUS与天花粉蛋白基因紧密连锁,PCR检测结果间接证明GUS基因也已导入小麦
【Abstract】 GUS and TCS (trichosanthin) genes were introduced into wheat via the pollen-tube pathway. Three wheat plants containing foreign TCS gene were detected from 125 plants tested. The transgenic plants were characterized by polymerase chain reaction analysis using two 24bp primers homolous to TCS gene. The result shows that the pollen tube pathway is suitable for genetic transformation of wheat cultivars.
