普萘洛尔及其对映体血药浓度对血浆心钠素水平的影响

【摘要】 本文研究8名健康志愿者口服单剂量普萘洛尔(40mg)和12名健康志愿者口服多剂量普萘洛尔(80mg/day×3)后;静息状态和踏车兴奋状态时血浆心钠素(ANP)水平的变化。口服单剂量普萘洛尔后3h,静息状态时血浆ANP水平升高19.4±11.1ng/L(P<0.05);而运动状态时血浆ANP升高更为明显,为64.8±40.9ng/L(P<0.05)。血浆ANP的变化与普萘洛尔对映体S(-)-PPL和R(+)-PPL浓度均具有良好的线性相关性,相关系数分别为0.8563和0.7192。口服多剂量普萘洛尔停药后12h,血浆ANP仍可维持较高水平,较基础对照值升高39.9±28.3ng/L(P<0.05)。此结果提示,普萘洛尔参与了血浆ANP水平的调控,这种作用尚难以用β肾上腺素能效应进行解释,可能与抑制ANP的体内降解过程有关。

【Abstract】 The aim of this study is attempt to elucidate whether the beta-adrenoceptor is involved in the regulation of the plasma levels of atrial natriuretic peptide (ANP) in & healthy volunteers who received a single dose of recemic propranolol (40mg) at rest and in 12 healthy volunteers who received multiple doses of racemic propranolol(80mg/day for 3 days ) during graded maximal ergocycle exercise.Plasma ANP concentrations were measured by radioimmunoassay.At 3h after oral single dose of racemic propranolol,the resting plasma ANP level was increased by 19. 4±11. lug/L compared with the hasal value (P<0. 05 ),and the exercising plasma ANP level was more significantly increased by 64. 8±40. 9ng/L (P<0. 05 ). There was significant correlation between the plasma ANP levels and the plasma ding concentlations of enantiomer S (-)-propranolol or R(+)-propranolol, the correlation coefficients were 0.9563 and 0.7192 respectively.At 12h after last dose in oral multiple doses of racemic propranolol, the rise was still maintained at a high levi, plasma ANP was increased by 39. 9±28. 3ng/L compared with the hial value (P<0. 05 ). These results suggest that propranolol is involved in the control of plasma ANP level, and beta-adrenoceptor mechanisms are not directly responsible for the regulation.
