具染色体不稳定特点的B淋巴母细胞株BCHI-1的建立
The establishment of B lymphoblastoid cell line characteristic of chromosome instability
【摘要】 经微量全血EB病毒转化法建立了一具染色体不稳定特点的B淋巴母细胞株(BCHI-1),该细胞株呈悬浮生长,核型为2n=46,XX。细胞群体倍增时间为52h。该细胞株表现为姐妹染色单体交换增高(16.88个/细胞),染色体畸变为4.5%,微核率25‰。该细胞株自发和丝裂霉素C诱发的染色体不稳性与正常的细胞株JMS95031相比均具有显著性差异(P<0.01)。
【Abstract】 A B-lymphoblastoid cell line (BCHI-1) characterized by chromosome instability was es- tablished by micro-whole blood EB virus transformation method. The results showed that this cell line was a suspension culture with normal karyotype 2n=46,XX. The mean cell population doubling time was 52 hours. BCHI-1 hnd elevated sister chromatid exchange (16.88 time/per cell),the chromosome abnormal rate was 4. 5%, micro nuclei rate was 25%o at normal condition. Both natural and induced chromosome instability of BCHI-1 had signficant difference from nor- mal B lymphoblastoid cell line (P<0.01).
