酿酒酵母和粟酒裂殖酵母属间融合和融合子特性
INTERGENERIC FUSION AND FUSANT CHARACTERISTICS OF SACCHAROMYCES CEREVISIAE AND SCHIZOSACCHAROMYCES POMBE
【摘要】 酿酒酵母(Saccharomyces cerevisiae Hansen)PW218和粟酒裂殖酵母(Schizosaccharomyces pombe Lindn)PW232的原生质体用20mmol/L CaCl2和30%PEG(MW6000)处理进行属间融合,获得了10多株融合子,融合率为0.65~1.96×10-5。对F2和F10两株融合子进行了葡萄糖、木糖及葡萄糖和木糖混合液的摇瓶实验结果表明F10融合子利用葡萄糖、木糖及两种糖混合液产乙醇的能力大大高于两亲株。F2融合子对木糖以及葡萄糖和木糖混合液的发酵能力亦较两亲株高,其中利用木糖产乙醇的量分别比PW218和PW232提高1.38倍和2.65倍。
【Abstract】 The protoplast of PW218 (Saccharomyces cerevisiae) was fused with the protoplast of PW232 (Schizosaccharomyces pombe) by 20 mmol /L CaCl2-30% PEG (MW6000) treatment. More than ten fusion strains was obtained by use of this method, and the fusion frequency was 0.65 - 1.96×10-5. Shake cultivations were performed for fusants F2 and F10 as well as their parental strains with xylose, glucose and mixture of xylose and glucose. The results show that F10 fusion strain has much higher ability of utilizing xylose, glucose and the mixture of xylose and glucose for ethanol production than its parental strains. F2 strain also has stronger fermentation capacity than its two parental strains in using xylose and the mixture of xylose and glucose. The ethanol productivity of F2 is 1.38 and 2.65 times as much as that of PW218 and PW232, respectively.
