颈总动脉的细胞间粘附分子表达和单核细胞粘附的免疫组化观察

【摘要】 细胞粘附分子与动脉硬化和缺血再灌流损伤等疾病有关.文内采用冷冻切片和免疫组化方法,观察比较了自发性高血压大鼠,具有中风倾向的自发性高血压大鼠和正常血压大鼠颈总动脉内皮的细胞间粘附分子表达及单核细胞与内皮细胞的粘附和聚集.结果显示:自发性高血压大鼠和具有中风倾向的自发性高血压大鼠颈总动脉内皮细胞有明显的细胞间粘附分子表达,而正常血压大鼠为阴性;与此结果相一致的是,自发性高血压大鼠和具有中风倾向的自发性高血压大鼠颈总动脉内皮粘附和聚集的单核细胞数也明显高于正常血压大鼠(3.0±0.5,5.3±0.9,和0;P<O.01).实验结果提示,高血压可能与颈总动脉细胞间粘附分子的表达及单核细胞粘附和聚集有关.

【Abstract】 The cell-adhesion molecules play an important role in the pathogenesis of atherosclerosis, ischemia/reperfusion injury and many other diseases. The cryostat sections and immunohis-tochemistry were used to investigate and compare the expression of intercellular adhesion molecule-1 (ICAM-1) and monocyte adhering to endothelial cells of carotid arteries in spontaneously hypertensive rats (SHR), stroke-prone SHR(SHR-SP) and normotensive Wistar-Kyoto (WKY) rats. The results showed that there was more CAM-1 expression on the endothelial cells of SHR and SHR-SP than that of WKY. Also, the numbers of monocyte adhering to endothelial cells were more in carotid arteries in SHR and SHR-SP than in WKY (3. 0±0. 5, 5. 3±0. 9 and 0 respectively), suggesting that hypertension might be associated with the expression of ICAM-1 on and increased monocytes adhering to endothelial cells of carotid arteries in SHR and SHR-SP.
