【摘要】 为探索经济有效的体外培养血管内皮细胞的方法,实验分别用(1)单纯胰酶;(2)全胶原酶:(3)胰酶(5份)和胶原酶(1份)三种方法分离牛主动脉内皮细胞.结果表明:用胰酶胶原酶混合消化的方式获取的内皮细胞的数量与全胶原酶获取的内皮细胞数量接近,而混合消化的方法大大降低了价格昂贵的胶原酶的用量,既经济效果又好.用相差显微镜、扫描电镜和透射电镜观察培养细胞的形态符合内皮细胞的特征.细胞八因子抗原染色阳性进一步证明培养的细胞是血管内皮细胞.更多还原

【Abstract】 We have developed an economical and effective method of collecting sufficient quantities of vascular endothelial cells. Vascular endothelial cells were successfully isolated by the combination of 0. 1 % collagenase and 0. 25% trypsin solution(1:5) from bovine aortic segments. They were demonstrated by factor VIII staining and proved to be of typical cobble-stone contact-inhibited morphology.更多还原
