逆转录病毒介导的β-珠蛋白基因在小鼠骨髓单核细胞的表达
EXPRESSION OF RETROVIRALLY TRANSDUCED HUMAN β-GLOBIN GENE IN MICE BONE MARROW MONONUCLEAR CELLS
【摘要】 从小鼠C57BL/6J股骨分离得到骨髓单核细胞(bonemarrowmononuclearcells,BMMC),并在体外建立了BMMC细胞培养体系。将携带有人β-珠蛋白基因及其增强子的逆转录病毒载体(N2AβE36)经DNA磷酸钙共沉淀转染单向性包装细胞系(ψ-2)收集抗G418阳性ψ-2细胞克隆的病毒上清,感染体外培养的BMMC,同时加入白介素-3(Interleukin-3,IL-3)、白介素-6(Interleukin-6,IL-6)、红细胞生成素(erythropoietin,Epo)等造血调节因子以促进造血细胞的增殖及提高基因转移的效率。经Southernblot和Northern-blot分析证实在小鼠BMMC中具有β-珠蛋白基因的整合与表达。
【Abstract】 Bone marrow mononuclear cells(BMMC)were obtained by flushing the femoral shafts of C57BL/6J mice and seperated by Ficoll.BMMCs were cultured in IMDM to establish the long-term culture. The vector(N2 AβE36)constructed with human β-globin gene and its enhancer was transfered into ecototropic packaging cell line(ψ-2)by calcium phosphate precipitation procedure. After selected with G418,the virus supertant of ψ-2 positive cell clone was collected and then infected BMMC.IL-3,IL-6,Epo etc,hematopoietic growth factors were added at the same time to stimulate the proliferation of BMMC and enhanc the efficiency of gene transfer.Southern blot analysis and Northern-blot analysis showed that human β-globin gene was integrated and expressed in BMMC.
