地鼠气管上皮损伤后粘液分泌细胞的修复作用
Repairing Action of Mucous-Secretory Cells after Injury of Tracbeal Epithelium in Hamsters
【摘要】 用改造后的静脉留置针将地鼠气管腹侧面约1/4的粘膜刮除,取材前1h腹腔内注入Brdu气管标本经HE、PAS,抗Brdu免疫染色,光、电镜观察。损伤后6h开始,边缘部上皮细胞增殖并向损伤面内爬行,胞浆呈PAS阳性,核呈Brdu阳性,24h覆盖损伤面。电镜下这些细胞表面无纤毛或仅有少量微绒毛,浆内有大小不等散在的粘液分泌颗粒和较多的粗面内质网。36h开始,损伤面由3一5层表皮样化生细胞构成,核周出现张力丝状物围绕,部分胞浆内仍有少量粘液颗粒。从72h开始,细胞出现短而细的纤毛或出现较多的粘液分泌空泡。2w后恢复正常气管上皮结构。这表明气管上皮中的粘液分泌细胞不是终末细胞,具有很强的DNA合成和增殖能力,是损伤后参与修复的主要细胞之一。
【Abstract】 bout one fourth of the tracheal mucosa of hamster was denuded by a venous needle reformed. Atone hour prior to killing each hamster was given a single intraneritoneal injecton of bromodeoxyuridineBrdu. tracheal specimens were stained with HE. PAS and anti-Brdu immunohistochemistry, then ob-served under the light and electron microscope. From 6h post injury the cells regenerated and migratedfrom the margin to wound site and the wound area was completely covered by a single layer of non-cili-ated flatted cells. Most of their nuclei were Brdu (+) and cytoplasms were PAS( + ) ,some mucous se-cretory granules and rich rER were observed. At 36h , 3-5 layers of epidermoid metaplasia cell: coveredall of the wound area and lots of tonofilament was observed around their nuclei. a few mucous granulesalso were seen in their cytoplasm. At 72h . there were short and small ciliates or many mucous granulesin theose cells. When 2 weeks the epithelium recovered the normal epithelial architecture. The resultsindicated that mucous secretory cells is not the end-stage cell . the ability to synthesize DNA and re-generate, but one of main cells of repairing tracheal epithelium after injury.
