测量农药和乙酰胆碱酯酶抑制物的光学式生物传感器
Optical Biosensor for the Determination of Pesticides and the Inhibitor of Acetylcholine Esterase
【摘要】 乙酰胆碱酯酶(AChE)是调节生物神经功能的一种酶,多种化合物,如有机磷和多种重金属离子,是乙酰胆碱酯酶活性的抑制成分。利用实验室自己合成的有色酶底物,2—(2—acetoxy—5—methyl—phenylazo)—N—methyl—1,3—thiazolium methosulfate(AMPT),它的溶液是黄色。在AChE酶的作用下,AMPT水解,其溶液转变为兰色的HMPT溶液(2—(2—hydroxy—5—methyl—phenylazo)—N—methyll,3—thiszokuym methosalfate)。但是,当有酶的抑制物质存在时,AChE酶会被抑制而失去部分活性。因此,溶液的颜色因酶抑制物质的浓度从兰色向黄色变化。利用光纤颜色测定仪测定溶液颜色的变化,就测定了抑制物质的浓度。
【Abstract】 Organo - phosphoric pestcides and some heavy mettalic ions are the inhibitor of acetylcholine esterase, so it can be detected by the defermination of the activity decrease of acetylcholine esterase. A novel optical fiber-based biosensor is reported based on the determination of the activity of acetylcholine esterase with the synthesized substrate of titled enzyme.
