用絮凝和超滤技术纯化木瓜蛋白酶
Purification of Papain by Means of Flocculation and Ultrafiltration
【摘要】 用无毒性的絮凝剂fc-l与无机助凝剂fc-2对木瓜乳胶水溶液预处理,然后用中空纤维超滤膜进行超滤浓缩。实验结果表明,用絮凝法预处理时,当絮凝条件为fc-l和fc-2各为0.2g/L,pH为5.0~5.5时,既解决了膜易污染的问题,又使木瓜酶得到了有效的纯化。在形成浓差极化-凝胶层的情况下,超滤速度随透出液体积和主体溶液浓度增大而减小,随料液错流速率的增大而增大,絮凝预处理有利于超滤速度的提高。
【Abstract】 Refining of papain by technology of flocculation and ultrafiltration are studied. The method is that pretreating the aqueous solution of papaya latex by use of the nonpoisonous flocculant fc-1 and the inorganic flocculant-aids fc-2, then extracting them by means of ultrafiltration. The experimental results show that 0. 2g/ L of flocculant fc-1,0.2g/L of flocculant fc-2 and pH 5.0~5.5 are the optimum conditions for flocculation. Ultrafiltration flux rate decrease with the increaments of permeate volume and bulk concentration, while it increase with the increament of fluid flow velocety acrossing the membrane.So flocculation will enhance the ultrafiltration flux rate of papain liquid.
