小麦根尖细胞分化过程中微丝骨架分布格局

【摘要】 以异硫氰四甲基若丹明-鬼笔环肽(TRITC-PHALLOIDIN)为探针,对小麦根尖三个不同发育区--分生区、伸长区和成熟区细胞中肌动蛋白纤丝(AFs,亦称微丝microfilament)的分布格局进行荧光显微观察和分析.结果表明:分生区细胞中细胞核为AFs网络包围,细胞周缘有AFS密集分布,胞质皮层中有AFs广泛均匀的分布;伸长区细胞中细胞核亦为网络包围,并明显可见AFs成束由核周围向四处辐射直到细胞周质,且与分布于细胞边缘的AFs汇集;成熟区细胞中伴随着核移向细胞的一侧,中央大液泡的形成,胞质皮层中只可见较微弱的荧光,但亦可见在核周围和细胞膜附近分布稍多.总之,随着小麦很尖细胞从分生到分化,细胞内AFs的分布格局发生了变化,且这种变化与细胞所处的状态及生理功能相关.

【Abstract】 he distribution of phalloidin labeling microfilaments has been examined during cell differentiation of wheat root tip. In meristematic cells there were a lot of microfilaments forming "baskets" around the nuclei,closely packed in the conical cytoplasm and fine peripheral networks close to the plasma membrance. In elongating cells,there also were microfilaments surrounding the nuclei, bundles of microfilaments radiating from nuclear "basket" and extending into the periplasm, and merging with micro filaments there. In mature cells,there were microfilaments aggregated around the nuclei,F-actin networks less densely distributed in the conical cytoplasm,and smaller amount of F-actin networks in the periplasm. In all,there were different patterns of microfilaments distribution in the root cells during differentiation.
