全反式维甲酸对转移性人肺癌细胞亚系恶性表型的影响

【摘要】 目的:研究全反式维甲酸(RA)对克隆化人肺癌高转移细胞亚系(PGCL3)恶性表型的影响。方法:在体外用5μmol/L和10μmol/LRA处理PGCL3细胞5d后,观察其对肿瘤生长、粘附、运动、侵袭能力和裸鼠体内实验性转移的影响。结果:上述肿瘤恶性表型指标均受到不同程度的抑制,其抑制作用在RA高浓度组更加明显。结论:RA可抑制高转移癌系的恶性表型,为RA治疗恶性肿瘤,特别是转移性恶性肿瘤提供了实验依据。

【Abstract】 ObJective:To find out the effects of all-trans retinoic acid(RA)on the malignant phenotype of a metastatic human lung cancer subline(PGCL3). MethOds:PGClt3 cells were pretreated with 5 μmol/L and 10 μmol/L RA in vitro for 5 days,then the effects of RA on in vitro growth,adhesive ca-pacity on laminin substrate,migrative and invasive ability(using Boyden chamber)of the pretreated PGCL3 cells were carried out in vitro,and the effects of RA on experimental metastatic ability of the pretreated PGCL3 cells vvas checked in nude mice. Results:The in vitro growth,adhesive capacity on laminin substrate,colony-forming ability in soft agar,migrative and invasive ability,and experimental metastatic capacity of pretreated PGCL3 cells were significantly inhibited.Conclusion:RA pretreatment could inhibited the malignant phenotype of a cloned metastatic human lung cancer subline,and the study may provide some reliable experimental data for the therapeutic use of RA on tumor invasion and metas-tasis.
