双向诱导H─2不同小鼠供受者间免疫耐受进行骨髓移植
TWO SIDES INOUCTION OF SPECIFIC IMUNOTOLERANCE BWTEEN H-2 MISMATCHED MICE FOR BONE MARROW TRANSPLANTATION
【摘要】 目的:预防骨髓移植后发生移植物抗宿主病和排斥反应。方法:紫外线B体内照射、紫外线B照射的供(受)者特异输血及环孢菌素A结合的方式、在供者(C57BL/6)、受者(Balb/c)小鼠间行双向诱导免疫耐受,再进行骨髓移植。结果:1只小鼠在移植后第30d死于移植物抗宿主病,9/10的小鼠存活>200d。而混合移植(Balb/c+C57BL/c→Balb/c,1:4)组,10/10的小鼠存活>200d。结论:双向诱导免疫耐受预防小鼠骨髓移植后发生移植物抗宿主病和排斥反应是必要的。
【Abstract】 Ohjectlve . in order to avoid both graft versus hoxt desease(GVHD(and rejection in hone niarrow transplantation(BMT).Methods. Ultraviolet B irradiation. ultraviolet B irradiated donor spe-cific transfusion (UV-DST. UV-RST)and CsA treatment were used in this study to induce antigen spe-cific tolerance between donor and recipient mice before BMT. kesults. 1 died on 30th day due to GVHD. while 9 survived >200 days without any evidence of GVHD. All of 10 mice who received mixed BMT(Balb/c+C57BL/6-Balb/c.1: 4) had lived > 200 davs. Conclusion . Two sides induction of speciflc immunotolerance between donorand recipient was necessary to prevent both GVHD and rejec-tinn for murine BMT.
