分子杂交技术检测免疫球蛋白基因重排的意义
DETECTION OF IMMUNOGLOBULIN GENE REARRANGEMkNT BY THE SOUTHERN BLOT HYBRIDIZATION
【摘要】 目的:探讨分子杂交技术检测免疫球蛋白基因重排的意义。方法:采用J_H和C_λ探针,经South-ern印迹法检测12例确诊或疑诊血液系统肿瘤患者的20份淋巴结、血和骨髓细胞的Ig基因重排。结果:发现B-NHL、CLL、HCL和MM均有不同于胚系的重排模式,而淀粉样变性、CML、T-ALL和鼻咽癌为胚系模式。结论:克隆性Ig基因重排仅见于B细胞,可作为B细胞克隆性增殖的证据,用于B细胞恶性增生性疾病的诊断,也可以用于分析B细胞肿瘤的克隆来源。
【Abstract】 bjective:To investigate the significance Clonal immunoglobulin gene rearrangement in lyrnpho node,blood and bone marrow of l2 patients with diagnostic or doubtful malignancy of hematopoietic system were detected by Southern blot hybridization using J_H and C_λ probes.Results:Of the 20 specimen,exa-mined,clonal immunoglobulin gfene rearrangements were found only in sample:of B cell malignancy,such as B-NHL,CLL,HCL and MM and not in non-B cell cancers,such as T-ALL,CML,Amyloido-sis and Nasopharyngeal carcinoma,ConclusiOns:The results indicate that detection of immunoglobulin gene rearrangement is a valuable method for diagnosis of various lymphoproliferative disorders and anal-ysis for clonal origin of B-cell tumor-
