镁钙离子诱导的人凝血因子Ⅸ、蛋白C的构象变化及其与相应特异单抗的连接状态
Experimental Study on Changes in Conformations or Factor Ⅸ and Protein C Induced by Metal Ions Mg ̄(2+) and Ca ̄(2+) and Their Connexion with Specific Monoclonal Antibodies
【摘要】 用圆二色光谱、荧光光谱及紫外差光谱研究镁钙离子诱导的FⅨ、PC的构象变化并通过蛋白负染电镜及荧光偏振实验观察了各构象与FⅨ、PC特异单抗的连接状况。FⅨ、PC在镁钙离子诱导下,其3种光谱均发生显著变化,并且与缓冲液中的金属离子浓度有关。FⅨ、PC的特异单抗仅与FⅨ、PC的某种特定构象发生连接反应。
【Abstract】 The changes in conformations of Factor Ⅸ and protein C induced by metalions Mg(2+) and Ca(2+) have been studied with CD spectra,fluorescence spectra and UV difference spectra,and the experimental observation on the connexion of all conformations with factor Ⅸ-and-protein C-specific monoclonal antibodies has been taken using protein negative stained electron microscope and fluorescence polarization,indicating that the significant changes in conformations of factor Ⅸ and protein C occured when being induced by the same ions,depending upon the concentration of the same ions in buffer and that the connexion of factor Ⅸ-and-protein C-specific monoclonal antibodies could take place only with some particular conformation of factor Ⅸ and protein C.
