不同培养基成分在球虫细胞培养中的作用
EFFECTS OF MEDIA AND THEIR COMPONENTS ON CELL CULTURE OF EIMERIA TENELLA
【摘要】 比较不同基础培养基(α-MEM、RPMI1640)和培养基的添加成分(胎牛血清、小牛血清、鸡 血清和胰岛素)对鸡球虫E.tenella(wis株)细胞培养的影响,结果发现α-MEM优于RPMI1640, 鸡血清优于国外普遍使用的胎牛血清。 胰岛素有促进细胞生长、促进球虫裂殖体和卵囊形成的作 用。
【Abstract】 The effects of media and their components,including different sera, Basic media,and insulin, were studied for in vitro culture of Eimeria tenella from sporozoites to oocysts. The growing speed of cells,the number of second generation schizonts and oocysts were taken as parameters for evaluation. The results indicated that chicken serum was better than fetal bovine serum and Q-MEM was better than RPMI 1640. Insulin in 0. 05μg/ml promoted either cell multiplication or the development of coccidia in vitro. It was suggested that chicken. serum should be used to cultivate Eimeria tenella after the chicken kidney cells were prepared in calf serum in order to increase the yield of oocysts. The functions of the components were also discussed.
