实验性动脉粥样硬化兔细胞膜Ca2+-ATP酶活性变化及药物对其活性的影响
Changes of Ca2+-ATPase Activity of Cellular Membrane in Experimental Atherosclerotic Rabbits and Effects of Drugs on Ca2+-ATPase Activity
【摘要】 观察了双侧髂股动脉去内皮及高胆固醇饲料喂养诱发的动脉粥样硬化兔细胞膜 Ca2+-ATP 酶活性的变化及药物穿心莲和鱼油对粥样硬化的预防作用及对 Ca2+-ATP酶活性的影响。结果表明,Ca2+-ATP 酶活性:粥样硬化组显著低于正常组(P<0.01)及两个药物处理组(P<0.05);血管造影见粥样硬化组双髂动脉均形成明显狭窄或闭塞,穿心莲提取物和鱼油均能明显降低狭窄发生率,减轻狭窄程度,尤以前者更明显。作者认为:由于某些因素使细胞膜 Ca2+-ATP 酶活性下降,导致细胞内 Ca2+浓度增加,从而促进血小板和单核细胞等分泌生长因子并促进血管平滑肌细胞向内膜移行和过度增生,形成粥样硬化病变。穿心莲提取物和鱼油能防止细胞膜 Ca2+-ATP 酶活性下降,因而能显著减轻粥样硬化病变。
【Abstract】 The changes of Ca2+-ATPase activity of cellular membrane in male rabbits with bilateral atherosclerotic ili- ac artery stenosis induced by deendotheliazation and high-cholesterol feeding were studied,and effects of Andro- graphis paniculata(AP)and fish oil(FO)on Ca2+-ATPase activity were observed.The results showed that Ca2+-ATPase activity in atherosclerosis group(AS)was significantly lower than that in normal control(P<0. 01).AP treated group and FO-treated group(P<0.05).Angiography showed that all rabbits in AS group had significant bilateral iliac stenosis or occlusion,while AP and FO.especially AP.could significantly reduce inci- dence of arterial stenosis and alleviate degree of stenosis.It was considered that cellular membrane Ca2+-ATPase activity reduction induced by some factors led to intracellular plasma Ca2+overload,the latter promoted platelets and m0nocytes to secrete growth factors and proliferation of vessel smooth muscle so as to induce formation of atherosclerosis.AP and FO could prevent the reduction of Ca2+-ATPase activity and therefore alleviate signifi- cantly arterial atherosclerotic lesions induced by both deendotheliazation and high-cholesterol feeding.
