通络熄风注射液对大鼠脑缺血再灌流损伤保护作用的实验研究

【摘要】 制备大鼠脑缺血再灌流模型,发现再灌流24h 后大鼠大脑皮质、海马、纹状体及血清中丙二醛(MDA)含量均显著升高,伴有脑匀浆乳酸脱氩酶(LDH)和磷酸肌酸激酶(CPK)活性的下降,血清 LDH、CPK 活性升高。采用通络熄风注射液防治组则 MDA 的升高受到明显抑制(P<0.01),同时脑匀浆中酶活性增强,血清中酶活性减弱(P<0.05)。中风指数评价显示通络熄风注射液防治组中风指数下降(P<0.01),并使脑缺血再灌流大鼠的神经病学症状和行为障碍减轻。从脑保护角度阐明了通络熄风注射液的临床药理基础。

【Abstract】 The protective effect of Tongluo-Xifeng injection(TLXF)on the free radical damage in cerebral ischemia/ reperfusion rats was reported,which was made by ironing 2 vertebral arteries and ligating 2 common carotid ar- teries for 25 minutes then reperfusing for 24 hours.The rats were randomly divided into 4 groups:10 animals for sham operation group;11 for large dose TLXF treated group.9 for small dose TLXF treated group;8 for nor- mal saline control group.The malondialdehyde(MDA)contents,lactate dehydrogenase(LDH)and creatine phosphokinase(CPK)activities in rats serum and brain homogenate was determined.The results showed that MDA contents of control group were more than those of sham operation group,while the MDA contents of TLXF group were less than those of control group in rats serum,cortex,hippocampus and striatum after 24 hours of reperfusion(P<0.01).LDH and CPK activities of control group were higher than those of sham opera- tion group and enzyme activities of TLXF group were lower than those of control group in rats’serum,the en- zyme activities in brain homogenate were elevated,but lowered in serum(P<0.05).In addition,the stroke in- dex of TLXF group was lower than that of control group.These results indicated that free radicals and lipid per- oxidation might be involved in cerebral ischemia/reperfusion injuries and TLXF would play an important role in protecting the brain from damage by free radicals.
