铅对肾上腺皮质激素生物合成毒作用的初步研究

【摘要】 多次给予豚鼠醋酸铅(30mg/kg)后,肾上腺皮质细胞脂质减少.部分细胞变性;血胆固醇降低而组织胆固醇在前几周降低,后期回升并有增加趋势;微粒体17a羟化酶在第2周升高.后期减少;血浆及肾上腺皮质醇水平在前3周升高.提示多次染毒早期.铅毒作用引起应激反应.导致一系列诸如胆固醇耗用过多、合成酶轻度诱导及激素水平增高,尿中代谢产物排出增加等改变。后期出现的改变,可能与肝脏损伤导致CBG减少、某些合成酶损伤有关。

【Abstract】 The toxic effect of lead on adrenocortical hormone’s biosynthesis was studied by using a subacute lead poisoning experiment of quinea pig. The results showed that, after multiple administration with Acetic Lead (30mg/kg )for four weeks,there was decreasing in the activity of microsomal cytochrome P450 172-Hydroxylase and in the content of cholesterol which was the precursor of adrenocortical hormone’s synthesis. No changes happened in the mitochondrial cytochrome P450 11βHydroxylase and P450 side-chain lyase. It was suggested that lead might have influence on some procedures of hormone synthesis. The increasing in the level of glucocorticoid of blood plasma and in the otput of urinary products of metabolism might relate to the stress reaction by lead,while the accumulation of hormones within adrenal cortex in the later stage might fesult from the decreasing of synthesis of corticosteroid binding globulin(CBG)manufactured by the liver.
