几种细胞分裂素诱导石竹外植体直接再分化成花芽的机理

【摘要】 用MS+2,4-D0.1mg/L为基本培养基,分别附加不同浓度的ZT、BA和KT,对石竹的叶片、茎和花等外植体直接再分化成花芽的试验表明:叶片的外植体在有BA时可再分化出只有红色花瓣的不正常花芽;在含ZT的培养基中再分化出具有1~2片叶或无叶的花芽,其花有两性花,雌花和仅单个雌蕊的不正常花,两性花可发育成结籽的果实;KT未能诱导花芽分化。茎外植体,3种CTK都不能诱导出花芽。花等外植体仅在KT3mg/L的培养中再分化出无叶的不正常花芽,ZT和BA的处理均不能诱导出花芽。

【Abstract】 Using the MS+2, 4-D 0. 1 mg/L as the basic culture medium to which different concenttation of zeatin, benzylaminopurine and kinetin were added. Direct differentiation of blade, stem and calyx explant to flower bud was studied. Blade explant could redifferentiate to red flower hod in medium containing benzylaminopurine 2 mg/L (Fig. 1). But in the medium containing zeatin 2 mg/L the blade explant redifferentiated to form flower bud with no leaf or one to two leaves.These flower budS could develop into hermaphroditic flower, pistillate flower or flower with single pistil.Hermaphroditic flower could form fruit which set seeds (Figs. 2~5 ). In medium containing Kinetin no flower bud was formed. The stem explants could not be induced to form flower buds in cyto kinin. The calyx explants redifferentiated to form flower buds with no leaf in medium containing kinetin 3 mg/L (Fig. 6), In medium containing zeatin or benzylaminopurine no flower bud was induced. The results show that the kind and dosage of hormone or growth regulating substance are dicisive factors in inducing flower differentiation. But the response to hormone or growth regulsting substance is different for different Parts of explants. On the other hand,if the blooming condition is given,the callus can by pass through the vegetative growing stage and form directly the reproductive sarge and differentiate to form flower bud with no leaf.
