应用PCR技术检测孕早期绒毛组织弓形虫DNA的研究

【摘要】 应用PCR技术检测270例孕6~12周妇女外周血及相应绒毛组织中的弓形虫DNA,结果弓形虫DNA阳性26例,相应的绒毛组织中检出8例,垂直传播率为30.77%(8/26)。同时用ELISA法检测血清中弓形虫循环抗原(CAg)、IgM和IgG抗体,结果阳性率分别为3.70%、7,03%和15.5%。IgM和/或CAg阳性者,外周血皆检出弓形虫DNA。提示应用PCR技术检测绒毛组织弓形虫DNA结合ELISA进行筛选,可早期诊断先天性弓形虫感染,较之用羊水、胎儿血进行产前诊断时间提前8~12周,有利于尽早治疗或中止妊娠。

【Abstract】 Polymerase Chain Reaction(PCR) was used to detect Toxoplasma gondii DNA(TOXO-DNA) in meternal blood and chorionic villus.A total of270 cases of pregnant women was surveyed.Circulating Antigen(CAg)、IgM. and IgG antibodies in sera were examined by ELISA in the mean time.The result showed that:(1) 26 cases of maternal blood TOXO-DNA were positive,including 8 cases of chorionic villus TOXO-DNA,The vertical transmission rate was 30.77%(8/26).(2) The positive rate of CAg was 3.70%,IgM7.03% and IgG 15.50%.(3) In case IgM and/or CAg were positive,TOXO-DNA could be detected in maternal blood.This study offers a simple,fast and accurat way for the diagnosis of congenital toxoplasmosis in the first trimester.It provides more clinical material for patient’s treatment.
