【摘要】 作者应用逆转录－多聚酶链反应（ＲＴ－ＰＣＲ）检测２５例急性白血病患者骨髓细胞中ｍｄｒ＿１基因的表达。以Ｋ５６２敏感细胞和体外诱导的耐药细胞株Ｋ５６２／Ａ０２分别作为阴性和阳性对照，发现复发和未缓解患者中ｍｄｒ＿１表达增高者占９０．９％（１０／１１），而初治患者中仅为２１．４％（３／１４），ｍｄｒ＿１表达增高者化疗效果差。应用单克隆抗体ＪＳＢ－１检测Ｐ－１７０结果与ＲＴ－ＰＣＲ一致。此ＲＴ－ＰＣＲ具有高度敏感性，可以检出２Ｐｇ的ｍｄｒ＿１ｃＤＮＡ，对于检测白血病ｍｄｒ＿１的表达是一种灵敏、特异的方法。更多还原

【Abstract】 y using a retrotranscriptase polyrnerase chain reac-tion (RT-PCR) technique, the expression of mdr_l genein 30 patients with leukemia was assayed. The mdr_lgene was specifically amplified by the PCR techniqueused. The mdr_l gene expressions in multidrug resistantcell line K562/A02 and its parental cell line K562 wereused as positive and negative controls, respectively. Themdr_l gene expression was detected in 90% (10/11) ofthe relapsed and refractory patients ,and only in 21%(3/14) of untreated patients. The high level expression ofmdr_l correlated with both the expression of P-glycopro-tein (P-170) and the resistance to chemotherapy. ThisRT-PCR method was highly sensitive and specific forthe assay of mdr_l gene in leukemia. The lowest limit ofsensitivity of this method was 2 pg of mdr_l cDNA.更多还原
