用逆转录-多聚酶链反应检测白血病患者多药耐药基因的表达
DETECTION OF MDR1 GENE EXPRESSION IN LEUKEMIAPATIENTS BY RT-PCR
【摘要】 作者应用逆转录-多聚酶链反应(RT-PCR)检测25例急性白血病患者骨髓细胞中mdr_1基因的表达。以K562敏感细胞和体外诱导的耐药细胞株K562/A02分别作为阴性和阳性对照,发现复发和未缓解患者中mdr_1表达增高者占90.9%(10/11),而初治患者中仅为21.4%(3/14),mdr_1表达增高者化疗效果差。应用单克隆抗体JSB-1检测P-170结果与RT-PCR一致。此RT-PCR具有高度敏感性,可以检出2Pg的mdr_1cDNA,对于检测白血病mdr_1的表达是一种灵敏、特异的方法。
【Abstract】 y using a retrotranscriptase polyrnerase chain reac-tion (RT-PCR) technique, the expression of mdr_l genein 30 patients with leukemia was assayed. The mdr_lgene was specifically amplified by the PCR techniqueused. The mdr_l gene expressions in multidrug resistantcell line K562/A02 and its parental cell line K562 wereused as positive and negative controls, respectively. Themdr_l gene expression was detected in 90% (10/11) ofthe relapsed and refractory patients ,and only in 21%(3/14) of untreated patients. The high level expression ofmdr_l correlated with both the expression of P-glycopro-tein (P-170) and the resistance to chemotherapy. ThisRT-PCR method was highly sensitive and specific forthe assay of mdr_l gene in leukemia. The lowest limit ofsensitivity of this method was 2 pg of mdr_l cDNA.
