新疆虫媒病毒的分离与鉴定

【摘要】 1990-1992年从新疆境内的多种婢、蚊、病人血清及脑脊液中分离到多株虫媒病毒,经初步鉴定,其中部分为甲属披膜病毒。1993年7~10月与美国国家疾病控制中心(CDC)虫媒传染病防治研究所合作,对上述病毒做进一步鉴定。35株病毒感染Vero细胞,其中28株可在3~7d内引起典型的细胞病变。用28株病毒进行空斑形成试验(PFT),其中19株可在Vero细胞中形成空斑,其滴度为log104.7/ml~log106.9/ml。对19株病毒继续做血清空斑减数中和试验(SDPRNT),其中91006与屈曲病毒(Chikungunya,CHIK)和都光和病毒(DoQuangHaT5-71,DQHT5-71)抗体中和滴度为2560,库孜拉加奇病毒(Kyzlagach,KYZ)抗体中和滴度为1280、盖塔病毒(Getah.GET)抗体中和滴度为320;90179与鹭山病毒(Sagiyama,SAG)抗体的中和滴度为1280,与CHIK、GET、KYZ和DQHT5-71抗体中和滴度均为640,与辛德毕斯病毒(Sindbis,SIN)抗体中和滴度为160;910160与CHIK抗体中和滴度为640,贝巴鲁病毒(Bebaru,?

【Abstract】 In the period from 1990 to 1992, we isolated. early or late. some arboviruses from pools of ticks and mosquitoes as well as from the samples of sera and cerebrospinal fluid obtained from some patients; of those viruses some were preliminarily identified as the members of Alphavirus. However. some viruses could not be identified into a species due to the limited conditions in our country. In the period between July and October 1993. we worked in collaboralion with the scientists from the Institute of Vector-Borne infectious Disease of the Centers for Disease Control. U.S.A. in making a further identification of the above mentioned viruses. The results showed that of 35 strains of the viruses with which Vero cells were infected, 28 strains caused a typical cytopathic effect between 3 and 7 days, and other 7 ones did not so. A plaque formation test was conducted in the 28 strains which had caused a typical cytopathic effect in Vero cells. The results revealed that of those strains 19 had a plaque formation in Vero cells; the plague titres being log104.7/ml-log106.9/ml, and the 9 remainders did not so. A serum dilution-plaque reduction neutralization test was further conducted in the 19 strains which had gave a plaque formation in Vero cells. The results showed that 15 of the 19 strains, except 4 strains (No.91006. 90179. 910160 and 92081) which gave a cross neutralization reaction with the antibodies against some viruses of Alphavirus. gave no obvious cross neutralization reaction with the antibodies against some tested viruses (CHIK, GET, SAG, SF. SIN, KY, DQHT5-71, DQHT9-71 and BEB, etc), indicating that these viruses might be some new arbovirus species.
