硒对培养人胚肝细胞Se-GSH-Px与MDA比值的影响

【摘要】 以白蛋白分泌能力及膜流动性为指标,观察原代培养人胚肝细胞经自由基攻击后的衰老变化及硒的影响。细胞共分4组:正常对照组,加硒对照组(预加硒),损伤模型组(加四氯化碳),硒防护组(加硒后加四氯化碳)。发现,损伤模型组细胞膜脂流动性明显下降,白蛋白分泌减少,伴随有丙二醛(MDA)浓度增高及硒谷胱甘肽过氧化物酶(Se-GSH-Px)活性降低,与正常对照组比较差异有显著性(P<0.001);加硒防护组上述损伤显著减轻(P<0.01),且其膜脂流动性与Se-GSH-Px/MDA比值呈显著正相关(r=0.9816,P<0.001)。结果表明,预加硒能提高细胞抗脂质过氧化能力,延缓细胞衰老。

【Abstract】 Abstract By taking the albumin secretion ability and membrane fluidity as indexes the ageing changes of primary cultured human hepatocytes after attacked by free radicals and the effect of selenium on them were studied.Primary cultured human fetal hepatocytes were pretreated with selenium and then exposed to carbon tetrachloride(CCl4).Malondialdehyde (MDA) levels and selenium dependent glutathione peroxidase(Se-GSH-Px) activity in the medium were determined. The cells were divided into 4 groups:normal control group,Se added control group.CCl4 injured group, and Se protected group(Se+CCl4).The results showed in the CCl4 injured group the membrane fluidity lowered markedly and albumin secretion decresed.together with increasing of MDA and lowering of Se-GSH-Px activity. There were significant differences as compared with normal control(P<0.001).In the Seprotected group the above mentioned damages were significantly less(P<0.01) and there was a significantly positive correlation between the membrane fluidity and the ratio of Se-GSH-Px/MDA (r=0.9816,P<O.001).These findings indicate that prophylactic supplementation of Se could enhance the ability of anti-lipoperoxidative damage in human hepatocytes and thus delay ageing.
