缺氧对肺动脉内皮细胞一氧化氮合成酶活性及基因表达的影响

【摘要】 以细胞化学技术和斑点印迹杂交对常氧和缺氧离体培养猪肺动脉内皮细胞一氧化氮合成酶(NOS)的活性和基因表达进行研究。结果显示:常氧培养肺动脉内皮细胞具有较高的NOS活性和信使核糖核酸(mRNA)转录水平;缺氧6、24小时后其活性和mRNA转录明显降低;缺氧48小时后其mRNA转录几乎消失;而无血清对照培养则对其活性和mRNA转录无明显影响。提示缺氧可能通过抑制肺动脉内皮细胞NOS的活性及其基因表达而在急性缺氧性肺血管收缩反应和慢性缺氧性肺动脉高压发病中起着重要作用。

【Abstract】 he aim of the study is to investigate whether hy-poxia roight affect nitric oxide synthase(NOS)activityor mRNA expression of cultured pulmonary artery en-dothelial cells in pigs by means of NADPH-diaphorasecytochemical stain and DNA-RNA dot blot hvbridiza-tion respectively. The NOS activity and mRNA expres-sion were highly present in nornioxic group as well as 6and 24 hours serum-free control groups,but significant-ly lowered in 6 and 24 hours hypoxic groups,The mR-NA expression of NOS gene was almost a bsent in 48hours hypoxic cells.The results indicate that hypoxiacan attenuate the activity and mRNA expression of NOSin cultured pulmonary artery endothelial cells, whichmay lay an important role in modulating acute hypoxicpulmonary vasocontriction and chronic hypoxic pul-monary arterial hypertehsion.
